Expression of the GUS Gene and Selection of Regenerated Rice Plants of MR81 Following Particle Bombardment

A reproducible system for producing hygromycin resistant rice plants of MR 81 was developed based on particle bombardment of embryogenic callus of mature embryos. Embryogenic callus was bombarded with the plasmid pRQ6 containing B-glucuronidase gene under the control of the CaMV 35S promoter using a PDS 1000 helium gun. Selection of 360 bombarded calli was carried out on hygromycin B. In six independent experiments, 9 calli differentiated into plants under continous selection of 50mg/l hygromycin B. Putative transofrmants were screened for GUS activity by the histochemical assay on cut leaf materials, and by growing the callus and regenerants on hygromycin selection media. Regenerated plants from the 9 selected calli survived under continous selection and showed GUS activity. [Authors' abstract].
