Modification of Mutagenesis Techniques to Enhance Isolation of High Producers of Citric Acid Mutants in Yarrowia Lipolytica

Colonies which exhibited significantly larger halo on the YEPG plates compared to the parent strain were isolated. Their acid production ability were reconfirmed on similar plates. Instead of growing isolated strains as shake flask cultures and assaying for their citric acid production capability, a more rapid method was developed. This involved removing agar plugs from the halo area of isolates grown on the YEPG plates using a cork borer. These plugs were then subjected to a 'freeae squeeze' regime to obtain samples for citric acid analysis by HPLC. [Authors' abstract].
