Cloning, Expression and Interaction Studies of the Potential RubisCO Activase CbbQ

Carboxysomes are polyhedral microcompartments found in all cyanobacteria and in many chemoautotrophs. Within their shell they contain the enzyme ribulose-1, 5-bisphosphate carboxylase/oxygenase (RubisCO), which fixes CO2. Downstream of the carboxysome operon there is another gene cluster containing several genes that may enhance carboxysome function. Two of these genes, cbbQ and cbbO, encode potential RubisCO activases. Using recombinant CbbQ and CbbO protein, and RubisCO isolated from carboxysomes the interaction between these proteins was studied. The CbbO and CbbQ proteins were both His tagged, allowing them to be purified with Ni2+-NTA column chromatography. Each of these tagged proteins was incubated with RubisCO on a Ni2+-NTA column to determine if both proteins could be co-eluted. These studies suggest that recombinant CbbO and CbbQ do not interact individually with RubisCO. Since His-tagged CbbO and untagged CbbQ, when co-expressed in E. coli, form a complex, ongoing studies are focused on determining if both potential activases are needed to interact with RubisCO when co-expressed. --Page iv.

Its core is issues which affect livelihoods of poor people in rural areas, but it also recognizes linkages - notably the role of non-natural resource components in livelihoods, the interests of the urban poor, and the role of associated factors such as health and education. It aims to provide topical analysis to inform development practitioners concerned with issues of poverty in development.

Design and Fabricate a Flexible Toilet Hose with Spool

This thesis explains the process of designing and fabricating of flexible toilet hose with spool. Firstly, the current toilet hoses are studied. The studies include the design, advantages, disadvantages, materials, and installation. Later, three concepts are suggested to be chosen and concept 3 is selected for further studies which involving three dimensional modelling using SolidWorks 2011 software. During the modelling process, the dimensioning of design product is decided. The fabrication method of flexible toilet hose with spool is later decided. The fabrication of this hose includes the process of shearing, cutting, roll bending, welding, drilling, grinding, filing, and finishing. The fabrication scale is 1:1 finely pilot design. The found out of problems also included. The problems covered from the start till project ends. Lastly, the conclusion of the project is documented and given some recommendation for further studies on the project.
